Example:

When we ask question to someones then we use Do, Does as helping verb before the subject.
Example:
1. Do you go to Hyderabad
2. Does she cook a meal
3. Does he play game
And in Negative tense we use Do not and Does not as helping verbs
Example:
1. I do not go to School
2. He does not go to Lahore
